New Protocol: Selinexor/pomalidomide/dexamethasone for relapsed/refractory multiple myeloma
Study:
  • Part of open-label Phase I/IIb STOMP – dose escalation and expansion phases
  • MM with >= 3 prior lines of therapy (to include PI and IMiD)
  • Selinexor (SEL) and pomalidomide (POM) dose escalated
Efficacy:
  • POM-naïve pts (n=33) => ORR: 58%, median PFS: 12.3 mos
  • Pts with previous POM (n=14): ORR: 36%, median PFS: 8.8 mos
Safety:
  • Grade 3-4 AEs: Neutropenia (56%), thrombocytopenia (35%), anemia (37%), fatigue (12%), nausea (2%)
  • Recommended phase II dose is SEL 60 mg PO wkly + POM 4 mg PO daily (21/28 d) + dexamethasone 40 mg wkly
